Position description
The successful applicant will work with a significant degree of
independence under the direct supervision of the PI in designing
and executing experimental studies in in vitro and in vivo model
systems, analyzing the data obtained from these studies, and
preparing/formatting these data in whatever context is needed for
their dissemination (e.g., publication, grant preparation, funding
report submission, etc.). Our research effort is focused on
understanding the gene-regulatory and epigenetic mechanisms that
govern early cell fate decisions during cardiogenesis and cell
state transitions in the setting of adult pathologic cardiac
remodeling. We have generated/are generating several novel lines of
mice and stem cells that harbor genetic alterations at specific
loci which will allow us to investigate the roles for a family of
histone acetyl-lysine reader proteins and nuclear tethering factors
in these processes. The candidate will require both theoretical and
hands-on skills in these areas of research in order to contribute
successfully to the growth of the lab. The successful candidate
will have the opportunity to eventually take on a lab manager role
with additional administrative responsibilities as well as
scientific and program management responsibilities.
Specific Job Responsibilities:
* Independently perform culturing and genomic manipulations of
mouse embryonic stem (mES) and human induced pluripotent stem
(hIPS) cells and directed differentiation to atrial and ventricular
cardiomyocytes as well as neural crest cells.
* Independently design, generate, and validate novel mouse genetic
alleles that selectively abolish specific functions of a family of
histone acetyl reader proteins or a transcription factor important
in resident fibroblast to activated myofibroblast cell state
transitions.
* Independently perform assessments of gene expression (from bulk
tissue and single-cells along with nascent transcription),
chromatin occupancy, and chromatin accessibility (from blk tissue
and single-cells) including sample collection and processing,
library preparation, multiplexed sequencing, and downstream data
analysis.
* Independently assess developmental consequences of
tissue-specific deletion of a family of histone acetyl reader
proteins or a transcription factor important in resident fibroblast
to activated myofibroblast cell state transitions.
* Independently perform standard molecular biology and biochemistry
techniques (including molecular cloning, quantitative reverse
transcription PCR, virus preparation and in vitro/in vivo viral
transduction, Western blotting, immunoprecipitation,
co-immunoprecipitation, and chromatin immunoprecipitation) as well
as high-resolution microscopy (including confocal microscopy and
live-cell imaging).
* Perform project management functions for large collaborative
projects involving multiple laboratories, oversee the day-to-day
tasks and responsibilities of a staff research associate (SRA), and
assume responsibility for maintenance of a 150-cage mouse
colony.
Required qualifications:
* PhD in Biological Sciences, Bioengineering, or related
fields.
* Advanced knowledge of stem cell culture (including mouse
embryonic and human induced pluripotent stem cells), standard
molecular biology and biochemistry techniques, next-generation
sequencing-based approaches to biological interrogations, and in
vivo mouse modeling.
* Demonstrated ability to contribute data and technical content to
grant proposals.
* Proven skills and experience in independently resolving broad
problems that may arise in collection or analysis of laboratory
data.
* Self-motivated with ability to work independently and as part of
a team. Able to learn effectively and meet deadlines.
* Demonstrated experience and ability to collaborate effectively
with all levels of staff; technical, students, faculty and
administrators.
* Demonstrated effective communication and interpersonal
skills.
* Demonstrated ability to communicate technical information to
technical and non-technical personnel at various levels in the
organization and to external research and education audiences.
* Proven ability to successfully work on multiple concurrent
projects.
* Demonstrate high integrity and professionalism to work with
patient data in a HIPAA-compliant and ethically responsible
manner.
* Ability to effectively manage own time.
Preferred qualifications:
* Advanced skills, and demonstrated experience associated with one
or more of the following: performance and analysis and
laboratory-based research including design, modification, and
implementation of genome-wide assays to interrogate consequences of
alterations in protein structure/function.
* Demonstrated ability to regularly interface with management.
* Interest and proven ability to train and guide the efforts of
junior members of the team towards carrying out the broader goals
of the laboratory.
* Track record of independent research.
* Completion of post-doctoral fellowship.

commensurate with the applicants experience and training.
